中文摘要: TGEV感染仔猪或体外接种细胞能显著诱导I型干扰素的的产生,借以发挥抗病毒作用。病毒感染过程中,p53可通过调节多种细胞信号转导通路影响病毒的复制和传播。项目组前期研究发现p53的早期活化抑制了TGEV的复制,那么在TGEV感染早期p53是否通过调控I型干扰素的产生及干扰素信号通路而发挥抗病毒的作用,目前尚未可知。本项目拟采用RNA干扰技术、荧光素酶报告基因试验、实时荧光定量PCR、激光共聚焦、EMSA及western blot技术等研究p53对TGEV诱生I型干扰素的调控作用,并深入研究p53调控TGEV诱生I型干扰素的分子机制,从而揭示p53通过调节I型干扰素介导的先天性抗病毒作用来发挥抗TGEV的作用,阐明TGEV感染早期p53抗病毒效应的分子机理。研究成果为以细胞信号分子为药物靶标进行抗病毒药物的研究提供思路和方法,为防治TGEV提供理论依据和帮助。

英文摘要: TGEV could induce the generation of type I interferon when infected piglet or inoculated cells in vitro, to play antiviral effect. p53 can regulate a variety of cell signal pathways to affect virus replication and spread during virus infection. Our previous research found that the early activation of p53 inhibit the replication of TGEV. It is unknown whether the p53 plays antiviral effect by influencing the generation of type I interferon and interferon signal pathways in early time of TGEV infection. In this study, the role of p53 in TGEV-induced type I interferon generation, and the molecular mechanism of p53 that affect the generation of type I interferon will be researched by siRNA, luciferase reporter gene test, real-time quantitative PCR, laser confocal microscopy, EMSA and western blot technology, to reveal the p53 could adjust the type I interferon mediated congenital antiviral function to play antiviral effect for TGEV, clarify the molecular mechanism of antiviral effect of TGEV during early infection. The results will provide new ideas and methods for the research of antiviral drugs through the cellular signaling molecular angle, provide theoretical basis and help for prevention and treatment of TGEV.
