中文摘要: 基于金纳米颗粒的侧向层析检测法因其快捷、简易和廉价而日益确立了在床边检测中的地位。目前,灵敏度和定量化检测已成为制约该检测法发展的两个重要因素。解决的关键在于增强金纳米颗粒的显色信号和对检测信号的高效收集。本项目利用金颗粒尺寸对其显色效应和光散射(表面增强拉曼散射效应,SERS)的影响具有一致性的特点,提出解决上述问题的新思路。研究在理论分析的基础上,拟构建一类普遍适用于侧向层析的、具有高效显色和SERS效应的双模传感性能并且抗体高度取向的免疫金纳米探针,以临床心脏标志物cTnI为检测目标,采用侧向层析的检测方式对cTnI进行基于聚集显色的高灵敏度检测,进而建立基于SERS光谱的强度与标志物含量的变化关系来实现检测结果定量化的方法,为进一步建立灵敏度高、检测结果可定量化的基于金纳米颗粒的侧向层析检测方法提供科学依据。

英文摘要: Gold nanoparticles based lateral-flow tests,due to their rapid,simple and low-cost characters, have become further established in point-of-care testing. Currently, the sensitivity and quantitative detection have become two important factor restricting the development of the lateral-flow assay.The key solution is to enhance the color signal of gold nanoparticles and efficient collection of the detection signal.The project takes advantage of the consistency of the size effects of gold particles size effects on their color sensing and light scattering characteristics (SERS), to propose new ideas to solve the problem.On the basis of theoretical analysis, this project tries to design a class of gold nanoparticles based immunoprobes with highly-oriented antibodies for colorimetric and light-scattering sensing high-sensitive Cardiac Troponin I by later-flow mode.Furthermore, we aim to establish a reliable method for quantitativly analysing test results based on the linear changes between in the intensity of SERS spectra and levels of cTnI markers. Implemeting this project can provide a scientific and application basis for high-sensitive, quantitative lateral-flow detection method based on gold nanoparticles.
