中文摘要: 心血管疾病标志物的检测已在心血管疾病的科学研究、临床诊治等领域发挥了重要作用。多组分心血管疾病标志物检测有助于心血管疾病的鉴别诊断、危险分层和预后判断。高灵敏心血管疾病标志物的检测方法,可实现对低浓度标志物的分析。.基于纳米材料信号放大和贵金属等离子体共振增强化学发光策略,本项目将检测抗体负载在多酶标记的树枝状DNA与银纳米粒子复合物的表面,以该纳米材料作为信号探针、以可抛式免疫传感器为基底,发展了一种可同时检测四种不同心血管疾病标志物的高灵敏化学发光图像免疫分析方法。.利用肌钙蛋白T(cTnT)、肌红蛋白(Mb)、肌酸激酶同工酶(CKMB)和C反应蛋白(CRP)作为模型分析物,该方法可实现对这四种不同标志物的同时测定。该方法达到了高通量和超高灵敏度的统一,为临床诊断提供了非常好的新方法。最终可延伸到中药成分与心血管疾病标志物的相互作用研究,在中药的等效成分群筛选中具有极大的应用前景。

英文摘要: The detection of cardiovascular disease markers has played an important role in the field of scientific research and clinical diagnosis and treatment of cardiovascular disease. Combination diagnosis of multiple markers contributed to the distinction and diagnasis, risk stratification and prognosis of cardiovascular diseases. High sensitive detection methods could realize the analysis of low concentration of markers..In views of nanomaterial-based signal amplification and metal plasmon resonance enhanced chemiluminescence, this item used the disposable immunosensor as the substrate, loaded the detection antibody (Ab2) on the silver nanoparticles (AgNPs) conjugated multienzyme labelled DNA dendrimer as the signal probe, and developed a high sensitive chemiluminescence imaging immunoassay method to detect four cardiovascular disease markers simultaneously..Using cardiac troponin T (cTnT), myoglobin (Mb), creatine kinase isoenzyme MB (CKMB) and C reactive protein (CRP) as model analytes, the method could be applied to detect four cardiovascular disease markers simultaneously. The method achieved the unity of high throughput and high sensitivity, providing a new tool for clinical diagnosis, and could be used to study the interaction between the component of traditional materia medica and cardiovascular disease markers. It has great application prospect in the screening of equivalent combinatorial components (BECCs) in traditional materia medica.
