中文摘要: 为了探明植物受铝胁迫诱导的基因及其作用和诱导机制,需要对植物抗铝毒进行基因表达调控的深入研究。本项目设计通过筛选拟南芥T-DNA突变体库。并从中获得了铝毒敏感突变体als1,通过质粒拯救和TAIL-PCR的方法获得T-DNA插入的基因组旁邻序列,测序结果表明此突变座位为MYB类转录因子。然后,通过遗传转化,获得了过量表达和功能互补的转基因拟南芥,利用这些材料,从生理生化及分子生物学等水平上对此转录因子基因进行功能分析。同时,进一步通过研究转录因子的相关基因上调表达分析,阐明调节抗铝毒基因表达的基因及分子机制。

英文摘要: To explore the Al stress-induced genes and their role, it needs to study the expression and regulation of aluminum tolerance genes. By screening Arabidopsis T-DNA mutant library, we gained an aluminum sensitive mutant als1, and got the flanking sequence of T-DNA insertion through the plasmid rescue and TAIL-PCR method. The sequencing results showed that the mutation site located in the MYB transcription factor. Then, through genetic transformation, we gained the excessive expression and the complementary function of transgenic Arabidopsis. By using these materials, we analyzed this transcription factor gene function at the level of physiological、biochemical and molecular biology. At the same time, by the expression analysis of relative genes, we further clarify the regulation of aluminum tolerance genes and their molecular mechanism.
