中文摘要: 有性生殖是高等植物生活史中的一个重要环节。其中,花粉萌发和花粉管极性生长是实现受精作用的关键,直接关系到受精过程能否顺利完成,然而目前对其发育与受精的分子机理尚不清楚。在本项目中,我们关注植物中研究较少、功能尚不清楚的HAP转录因子,以我国重要的特色树种-青杄为试材,利用花粉体外培养、基因枪瞬时转化、RNA interference等技术,结合激光共聚焦扫描显微镜观察,从分子、细胞等水平研究HAP转录因子在花粉萌发和花粉管发育过程中的作用和功能;利用酵母单杂交、染色质免疫共沉淀技术结合Pulldown等蛋白互作研究手段,筛选HAP3和HAP5亚基调控的下游靶基因及其作用的分子机制等,为揭示HAP家族转录因子在植物体内的功能及其参与的植物有性生殖过程的转录调控网络提供重要基础。

英文摘要: Plant sexual reproduction is one of the key processes during life history in higher plants. Among them, pollen germination and pollen tube growth are vital to double fertilization. However, relatively little is known about the biological function of HAP complex in plants compared with its role in yeast and mammals. In the project, we are going to investigate the role and function of HAP transcription factors in plant sex reproduction process from the molecuar and cellular levels using in vitro culture of pollen,microprojectile bombardment,RNA interference technique in combination with confocal laser scanning microscopy and screen the target genes based on the yeast one-hybrid and ChIP technique. The results would be of importance for researchers to understand the role and function of HAP family transcription factors and the molecuar mechanisms of HAP invovled in plant sex reproduction process.
