中文摘要: 蠕虫形粒子在生物医药领域具有显著不同于球形粒子的性质,如较长的血液循环时间、较低的巨噬细胞内吞率或较高的肿瘤细胞摄入率。在线形聚合物主链上高密度接枝线形聚合物支链,所得的聚合物刷是一类结构稳定、尺寸均一的单分子蠕虫形纳米粒子,如能方便地调控粒子的功能性,则非常适合作为模型研究非球形粒子的生物学性质。本项目拟研究通过侧链端基修饰或引入聚电解质侧链的方式对亲水性蠕虫形聚合物刷进行功能化的普适方法。利用高效偶联反应,可以得到侧链端基表面带有羧基、氨基、羟基、磺酸基、糖基等大量官能团的聚合物刷;将聚电解质链按一定比例接入聚合物主链,可以得到侧链区带有正/负电性的聚合物刷,灵活简便地实现对水溶性蠕虫形聚合物刷不同结构区域的功能化。通过功能性蠕虫形粒子与多种细胞相互作用行为及其对基因或药物运输能力的研究,初步揭示纳米粒子形状及功能化所带来的生物学特性。

英文摘要: Wormlike nanoparticles possess special properties in biological and medical fields. Compared to spherical particles, they have prolonged blood circulation time, less endocytosis by macrophages and more intenalization within cancer cells. Polymer brush with high density side chains displays wormlike morphology, which is an ideal model for biological researches of non-spherical particles. This program is planed to study on the functionalization of wormlike polymer brushes by modifying end groups of the side chains or by introducing poiyelectrolyte side chains. A variety of functional groups, such as carboxyl group, amino group, hydroxyl group, sulfo group and glucose units, will be modified onto the surface of the polymer brush. Poiyelectrolytes will be grafted onto the polymer backbone in fixed proportion. The obtained wormlike polymer brush will be water-soluble, and will express functionalities in different structural regions. The interactions between wormlike brushes and cells, as well as the gene and drug delivery abilities of the functionalized brushes, will be studied on. The biological characterstics caused by the shape and functionalization of the nanoparticle will be revealed,
