中文摘要: T细胞受体(TCR)是T细胞表面识别抗原的分子。人体免疫系统具有一个庞大的T细胞受体库,通过VDJ重组,人体的T细胞受体多样性的数目超过一千万。近年来兴起的高通量测序技术可以一次性测出上百万个TCR分子,为全面刻画T细胞受体库特征提供了契机。肿瘤浸润T细胞是抑制肿瘤和过继免疫治疗的的重要免疫效应细胞。本项目拟以直肠癌患者的肿瘤浸润T细胞为研究对象,将采用深度测序方法对人体T细胞受体库多样性进行深入研究。并且结合生物信息学的分析,分离出其中大量表达的T细胞受体,以此来筛选肿瘤抗原特异性T细胞受体。将T细胞受体库的多样性和病人的临床表现进行关联,提出一个临床预测的方法,为免疫特异性治疗提供研究基础。

英文摘要: T cells recognize antigens by T cell receptors (TCR). Due to genetic recombination of germline cells, the diversity of TCR repertoire is estimated to be more than 10 millions. Recently, high throughput sequencing technique has been applied to sequence millions of TCRs at a single run, which enables us to investigate the global features of immune repertoires. Tumor infiltrating T cell (TIL) is a key player in restrcting tumor growth and in immunotherapy. In this project, we propose to study the diversity of TCR repertoire in colorectal tumors by high throughput sequencing. We aim to identify the tumor antigen specific T cell receptors by selecting highly expressed T cell receptors, based on bioinformatic analysis. By correlating the diversity of TCRs with patients' clinlic outcomes, this study will develop a prognostic tool in the treatment of colorectal cancer and possibly other malignancies.
