中文摘要: 猪传染性胃肠炎病毒(TGEV)感染引起各年龄段猪出现急性腹泻和严重脱水,但TGEV的致病机理尚不清楚。通过一系列研究,我们首次发现TGEV感染猪小肠上皮细胞(SIEC)后,活性氧(ROS)和硫氧还蛋白的表达水平明显升高,初步表明TGEV感染引起SIEC细胞发生氧化应激(OS)。但TGEV引起OS的具体机制尚不明确。因此,本项目在确证不同宿主细胞内TGEV感染和OS的关系基础上,进一步寻找引起OS的关键病毒蛋白,揭示氧化应激对宿主细胞生理功能的影响,并通过探明OS下游信号蛋白的变化深入揭示TGEV引起氧化应激的分子机制,同时应用添加抗氧化剂、RNA干扰和过表达三种方法综合探明氧化应激对TGEV病毒复制的影响,研究获得的成果将为揭示猪传染性胃肠炎病毒的致病机制提供新的科学资料。

英文摘要: The infection of transmissible gastroenteritis virus (TGEV) could cause contagious viral disease of all aged pigs, characterized with severe diarrhea, and dehydration, however, the pathogenesis of TGEV is not clear by now. We firstly found that the level of relative oxygen species (ROS) was significantly elevated and the mRNA level of thioredoxin (Trx) was increased in the TGEV infected swine intestinal epithelial cells (SIECs), which indicated that OS might play an important role in the pathogenesis of TGEV. Whereas the mechanism needs more explored. In this present study, the effect of OS induced by TGEV will be identified on IEC cells, ST cells and PK-15 cells. The key viral proteins in inducing OS is about to be detected and the influence of OS on cell cycle and apoptosis will be verified. Furthermore, the molecular pathway of OS is expected to be elucidated by detecting the downstream proteins. Besides, the exact roles of OS in the TGEV replication could be detected by adding the antioxidants, RNA interference and over expression of antioxidant proteins in the TGEV infected cells. Taken together, these findings will provide new evidences to the pathogenesis and mechanism of TGEV.
