中文摘要: BTF广泛存在于哺乳动物细胞中,是重要的促凋亡和转录抑制因子。研究表明,BTF在多种亚型的疱疹病毒感染过程中都具有抗病毒活性;相应地,BTF也成为病毒编码蛋白攻击的靶点,受到抑制和降解。但BTF的抗病毒机制及相应的病毒逃逸机制仍不清楚。目前的研究主要集中在感染人的病毒,对BTF抵抗其他动物病毒的研究尚未见报道。我们前期研究发现 BTF在猪伪狂犬病毒( PRV)的早期感染中受到显著抑制,提示BTF参与到宿主细胞对猪疱疹病毒的内在防御机制中,同时病毒也通过抑制BTF的方式逃逸其抗病毒作用。本研究拟采用细胞分子生物学、生物化学和病毒学等方法,研究BTF与PRV之间的相互作用和功能关系。具体内容包括:1)研究BTF对PRV的抗病毒能力和作用机制;2)探索PRV对BTF的影响及逃逸机制。本研究将增加我们对BTF抗病毒生物学功能和分子机制的认识,并为动物疫病的预防和控制提供重要的理论基础和实践指导。

英文摘要: Apoptosis-associated protein BTF was identified in most mammalian cells. Initial studies indicated a role for this protein as an inducer of apoptosis and repressor of transcription. Subsequent studies have shown that BTF plays critical roles in the antiviral defenses of host intrinsic immune response, especially in the infection process of multiple human herpesviruses. In contrary, viruses have evolved different ways to counteract BTF-mediated cellular antiviral responses, such as the degradation and repression of BTF protein. To date, the molecular mechanisms of the antiviral actions and its corresponding escape are still unclear. Our previous studies demonstrated that BTF protein was dramatically decreased in the early stage of porcine pseudorabies virus (PRV) infection, suggesting the important roles of BTF in antiviral defenses of PRV. In this proposal, taking porcine pseudorabies virus infection as a model, we will elucidate the interplay of BTF with PRV in infection to reveal the important roles of BTF in antiviral defenses and the corresponding escape mechanisms of viruses. Specifically, we will 1) investigate the potential antiviral activities of BTF in PRV infection and explore the mechanisms responsible; 2) determine the regulation and escape mechanisms of PRV to BTF. The results coming from this proposal will greatly enhance the understanding of biological function and molecular mechanisms of BTF in antiviral defense, which would provide important theoretical basis and practical guidance for prevention and control of animal diseases.
