中文摘要: 口蹄疫 (FMD) 是由FMD病毒(FMDV)引起的牛、羊等发生的一种烈性传染病,严重危害畜牧业的发展。由于FMDV血清型多且变异快,不同血清型疫苗间无交叉保护,通过疫苗免疫的单一手段很难防控FMD。因此,科学家们开始关注病毒感染机制,尤其是病毒关键性受体的研究,以期通过封闭或敲除受体基因阻断病毒感染,为控制和根除FMD提供新思路。体外研究表明,整合素β#20122;基是FMDV整合素受体中与α#20122;基配对的最主要的β20122;基,但是否是体内关键性受体亚基,尚待验证。本研究用FMDV接种3日龄β/-及β/+乳鼠,前者可完全抵御低剂量及部分抵御高剂量FMDV的攻击,而后者全部发病且多数死亡。利用RNAi部分沉默β/-乳鼠的整合素β#25110;β#20122;基基因表达后,乳鼠抵抗高剂量病毒攻击的能力明显增强。本课题利用转基因乳鼠模型首次体内验证了β#26159;FMDV感染的关键性受体亚基,并探讨了整合素β#21463;体亚基与其它亚基在病毒感染过程中的协同作用,为深入研究整合素β#20122;基在FMDV感染天然易感动物中的作用研究奠定了基础。

英文摘要: Foot-and-mouth disease (FMD) virus (FMDV) infecting cattle, sheep and other animal caused a infectious disease, which occurred in a serious harm to the development of animal husbandry. There are more serotypes, quick mutation of virus, and no cross-protection among vaccines against different serotypes, so only vaccination is too difficult to prevent and control FMD. Therefore, the scientists started to pay attention infection mechanisms, in particular the study of key receptor for the virus entry, once it is blocked or knockout, viral infection may be stopped, so it will be a new strategy for control and eradication of FMD. In vitro studies have shown that integrin βsubunit is the major βubunit of integrin αsubfamily as a receptor of FMDV, but it is not clear whether it is the key βubunit in vivo.In this study, 3-day-old β/- and β/+ suckling mice were inoculated with FMDV, β/- mice can be completely against low-dose and partly against high dose of FMDV, whereas all β/+ mice were morbid and the majority of them were mortal. After integrin βor βgene was partly silent with specific RNAi, the β/- suckling mice could more resist against high-dose of FMDV. It is the first time to confirm that integrin βgene is the key βubuni of receptor of FMDV infection with transgenic mice, and to explore the synergy between integrin βand other βubunits, it will be basis for further assessing the role of βsubunit in FMDV infection of natural sensitive animals.
