中文摘要: 肝癌发病率存在明显的性别差异:男性发病率是女性的3-5倍,提示肝癌的性别差异可能与性激素在肝癌形成中的作用有关。目前认为,性激素,特别是雌激素,在肝癌发生中的作用存在两面性。硫酸基转移酶SULT1E1是一种参与雌激素代谢的关键酶,可通过硫酸化作用而使雌激素灭活。本课题组在肝癌基因表达谱芯片数据分析中发现,SULT1E1在肝癌中的表达显著降低。预实验结果也发现,过表达SULT1E1可显著抑制肝癌细胞系的生长,初步结果提示SULT1E1可能在肝癌的发生中起一定作用。那么SULT1E1是否在肝癌发生及进程中发挥关键调控作用?其作用方式是否与雌激素有关?本课题旨在探讨SULT1E1在雌激素对肝癌生物学行为的影响中发挥的作用及其机制,并探讨其作为肝癌临床诊疗靶标的可能性。该课题的完成将有助于深入理解肝癌发生发展与激素作用的相互关系,同时为肝癌的新型临床诊疗手段的研发提供确实和重要的理论和实验依据。

英文摘要: The hepatocellular carcinoma (HCC) is more frequent in men. The incidence is 3-5 times that of women, suggesting that sex hormones may play important roles in the tumorigenesis in HCC. Sulfotransferase family 1E (SULT1E1) is the major sulfotransferase isoform responsible for the inactivation of beta-estradiol via sulfation at physiological concentrations, therefore its role in altering liver estrogen levels may be significant in the onset and progression of HCC. Previously we have observed the down-regulated expression of SULT1E1 in HCC tissues, and that SULT1E1 overexpression significantly inhibits the proliferation of liver cancer cells. Here we attempt to investigate the potential roles and the underlying mechanisms of SULT1E1 in the effects of estrogen on HCC behaviors. The work will facilitate the comprehensions of the relationships between estrogen and the tumorigenesis in HCC, and provide theoretical and experimental basis for the development of novel diagnostic and therapeutic approaches of HCC.
