中文摘要: 瓠瓜(Lagenaria siceraria)是瓜类植物常用的一种较为理想的砧木材料。由于嫁接苗既保持了接穗品种的优良性状,又利用了砧木的有利特性,从而能够达到改变植物的一些生长特性如开花、结果、抗性甚至植物形态的目的,因此,嫁接成为植物防止土传病害、提高产量、增强抗逆性的一种简便有效的途径。而全面深入地了解植物嫁接过程中砧穗互作机制是揭示嫁接对植物生长发育及形态变化影响的必要前提。因此本项目拟通过双向电泳技术寻找嫁接后瓠瓜砧穗互作产生的特异表达蛋白,并通过高通量质谱鉴定技术获得差异点蛋白的EST序列,以此设计引物,分离克隆与瓠瓜嫁接互作相关的基因,并通过荧光定量表达分析这些差异基因在嫁接互作中的表达模式,从蛋白质水平到基因水平的视角上了解它们在嫁接过程中的相互作用关系和遗传信息交换机制,进一步了解植物嫁接互作的实质。

英文摘要: Bottle Gourd (Lagenaria siceraria)as an ideal stock is commonly used in Cucurbitaceae. Grafting is an effective way to prevent wilt and other soil-borne diseases, increase yield, enhance resistance in plant breeding because Grafted plant capable of the benefits of certain scion cultivars combined with certain rootstocks , can change some characteristics of plant development including flowering、fruiting、resistance and even plant morphology. In order to reveal the influence of grafting on plant growth and morphological changes,The project intends to identify differential proteins and its EST sequences bewteen rootstock and scion in the grafting of bottle gourd by two-dimensional electrophoresis and high-throughput mass spectrometry; then clone the genes with the primers designed from the known EST sequences identified from the differrential proteins and analysis those gene expression patterns by means of real-time fluorescence quantitative.The aim of the research is to understand the interaction between rootstock and scion and the genetic information exchange mechanism in the grafting process from protein level to gene and learn more about the essence of grafting mechanism in plant.
