中文摘要: 空气动力学直径小于2.5μm的大气颗粒物(PM2.5)对人体健康危害重大,但目前尚未阐明PM2.5导致人体健康影响的效应机制及关键物种。作为PM2.5的重要生物组分,内毒素暴露能诱发一系列的健康效应,但相关研究较少。本项目拟对北京市实际大气细颗粒物一年的连续观测样品中的内毒素及其它化学组分对细胞产生的包括氧化损伤、炎性因子、DNA损伤、细胞凋亡/死亡在内的毒性效应进行测量,通过①统计各组分与细胞损伤程度之间的相关性,②比对样品和等量标准品对细胞损伤的程度,③观察定向抑制某组分后剩余颗粒物对细胞损伤的程度,综合比对分析三部分结果,旨在研究内毒素对PM2.5的毒性贡献及与其它组分的协同作用,完善颗粒物健康效应机制。本项目为颗粒物暴露研究提供了更多的视角,通过区分不同组分对PM2.5毒性效应的影响,探索关键效应物种,推测颗粒物健康效应机制,为我国大气污染源控制和公共卫生干预政策提供理论依据。

英文摘要: Exposure to airborne particulate matter with an aerodynamic diameter of less than 2.5μm (PM2.5) is the most health-damaging environmental factor to human. However, understanding the biologic causal mechanism and essential species of the PM2.5 health effect remains challenging. As an important biological component of PM, ambient airborne endotoxin reaches high concentrations, and inhalation of endotoxin is reported to induce acute and chronic health effects. Despite this, however, endotoxin is less studied. In this project, one year continuous measurements of endotoxin and other chemical components in PM2.5 will be performed. Also, their contribution to the functional capacity modulation of human alveolar epithelial cells will be assessed, including oxidative potential, inflammatory factors, DNA damage, apoptosis and cytotoxicity. Statistical analysis, standard sample comparisons and inhibition of special components will be performed to find out the toxicity contributions, synergistic effects and effect mechanisms of endotoxin and other chemical components of PM2.5 in different months throughout one year. We hope to complement the biologic causal mechanism and essential species of the PM2.5 health effect through the project, supporting the pollution source control and public health intervention policy.
