中文摘要: 生物膜富含信号分子,是感知和传递外界信号的重要部位,磷脂酶D(phospholipase D, PLD)通过水解磷脂而产生信号分子磷脂酸(phosphatidic acid, PA),并导致生物膜系统的重组、膜上信号分子的移位及其活性变化。近年来的研究结果表明,植物的磷脂酶D 比动物的更为多样性和复杂性,参与了多种生物过程。 最近,申请人在模式植物拟南芥中鉴定了一个新型的磷脂酶PLDepsilon(PLDe),并发现其参与氮信号及植物生长调节的过程。本研究将在前期研究的基础上,采用分子遗传、生化、代谢分析及信号转导等方面全方位探讨PLDe 在氮吸收、同化和植物生长中的作用及其机理,并通过蛋白组学、免疫学、质谱分析等手段探明PLDe 介导的氮素同化和生长的下游靶标分子及其信号通路,从中进一步鉴定出对氮感应和植物生长起促进作用的关键因子,为作物氮高效利用和高产育种奠定基础。

英文摘要: Cell membranes serve as an initial and focal point of signal perception and transduction. Phospholipase D (PLD) hydrolyzes membrane phospholipids to produce phosphatidic acid (PA), an important signal molecule in cells. The activation of PLD results in membrane remodeling, re-localization and activity changes of effectors associated with membranes. Plant PLD is much more complicated than that of animals, which is involved in various biological processes. Recently, I have identified a novel PLD, PLDepsilon (PLDe). Preliminary study indicates that PLDe is involved in nitrogen response in Arabidopsis. This study will further characterize the function and mechanism of PLDe in plant growth in response to nitrogen availability via the combination of genetic, biochemical, and metabolic analyses, as well as signaling transduction. In addition, the downstream target(s) of PLDe will be identified to reveal the pivotal factor in N sensing and plant growth. The completion of study will provide molecular basis for developing high yield of crop plants.
