中文摘要: 木尔坦棉花曲叶病毒(CLCuMV)是严重威胁棉花生产的双生病毒,其编码的βC1蛋白涉及病毒运动和症状。为了理解CLCuMV侵染的机制,我们用CLCuMV βC1做诱饵筛选酵母双杂交文库,发现植物蛋白SKP1与CLCuMV βC1互作。我们利用双分子互补实验发现βC1与SKP1在植物体内的互作,初步研究发现SKP1基因沉默可导致CLCuMV的侵染力下降,βC1的表达能抑制受泛素化调控的茉莉酸(JA)及水杨酸(SA)信号途径,体外喷施JA和SA能够减缓病毒的侵染。本课题将进一步研究SKP1与βC1互作的定位、互作以及植物泛素途径在CLCuMV侵染中的作用,我们的研究可加深对病毒侵染机制的理解。

英文摘要: Cotton leaf curl Multan Virus(CLCuMV) is the major threat to cotton production. CLCuMV-encoded βC1 is involved in virus movement and symptom. In order to understand the mechanism of plant virus infection. We screened yeast 2-hybrid library by using CLCuMV βC1 as a bait, and found that CLCuMV βC1 interacts with SKP1. Our preliminary data suggests that CLCuMV βC1 interacts with SKP1 in vivo, and silencing of Skp1 reduces CLCuMV infection. CLCuMV βC1 can interfere Jasmonic Acid and Salicylic Acid pathway. Spraying exogenous JA or SA can attenuate CLCuMV infection. This study will investigate the role of interaction of CLCuMV βC1 with SKP1, and the role of ubiquitnation in virus infection. This study will help to understand how virus infects plants.
