中文摘要: 细菌骨架蛋白MreB是真核细胞肌动蛋白Actin的同源蛋白。在细胞内,MreB聚合为特异的动态结构,作为组织中心提供细胞及亚细胞结构形成的力、模板和方向,参与多种细胞过程。细胞作为一个整体,在细胞周期的任一时刻,以MreB为模板或支架的生物大分子的空间结构和分布之间都会关联和相互影响。遗憾的是,目前已有的数据都是对MreB及其某个特定作用蛋白间的结构和功能的孤立研究,而对MreB与多个作用蛋白间结构和分布在空间位置上的联系以及相应的功能上的协调还没有研究。 我们将利用超高分辨率荧光显微镜结合分子生物学技术研究MreB及与其作用的多种蛋白质的空间位置和分布,建立一个以MreB为核心节点的空间网络的动态高分辨率图像。这个层面的结果将使我们可以更全面地理解MreB在细胞周期的各个时刻是如何通过其动态结构来发挥组织和调节功能的,并以此为模式来回答细胞构造与遗传信息及代谢等细胞过程是如何联系起来的。

英文摘要: Bacterial cytoskeletal protein MreB is the homolog of eukaryotic cytoskeletal actin. In the cell, MreB polymerizes into certain dynamic structures, which, as organizing centers, provide force, templates and guidance for the formation of cell architecture and subcellular structures. A cell being an integrative entity, at any moment of its cycle, spatial structures and distributions of macromolecules that use MreB as templates and scaffolds may interact with each other. Unfortunately, by far most studies are isolating as they have been focused on the structures and fuctions of MreB and one particular MreB-interacting protein at a time. There have been lacking systematic studies on the connections among structures formed by MreB and multiple proteins, neither the coordination of their functions. In this proposal, we are going to combine super-resolution fluorescence microscopy with molecular biology techniques to study the spatial location and organization of MreB and multiple MreB-interacting proteins. With that, we hope to constuct a dynamic, super-resolution image of the spatial network organized around MreB cytoskeleton. Results at the network level will help us understand more globally how MreB uses its dynamic structures to perform the organizing and regulating functions at different moments of the cell cycl
