中文摘要: 缺血性心脏病的防治已成为社会及医学界共同关注的焦点,寻找新的心脏保护机制及防治药物靶点已成为人们关注的重要课题。本课题基于药物预处理,以线粒体ATP敏感钾通道(mitoKATP)为切入点,采用体内体外缺血再灌注模型,研究心肌缺血状态下mitoKATP功能、电生理学特性的变化规律,并观察缺血再灌注不同阶段自噬水平的变化,研究自噬在mitoKATP开放剂及阻断剂预处理心肌缺血再灌注损伤中的作用。该研究将揭示心肌缺血缺氧时mitoKATP功能、电生理学特性的变化规律,明确mitoKATP所处的地位,以及自噬在mitoKATP开放剂预处理心肌缺血再灌注损伤中的作用。阐明mitoKATP开放剂预处理对心肌缺血的保护作用,证实mitoKATP开放诱导自噬水平增加并通过磷脂酰肌醇3-激酶信号通路产生心肌保护作用,为临床防治心肌缺血及再灌注损伤提供新的干预手段。

英文摘要: Prevention and treatment of ischemic heart disease has become the common focus of the social and the medical profession. Therefore, looking for a new protection mechanism and drug targets has become an important issue. This subject is based on pharmacological preconditioning, with Mitochondria ATP sensitive potassium channel (mitoKATP) as the starting point, and discusses the changes of functions and electrophysiological properties of mitoKATP and the level of autophagy during myocardial ischemia by using in vitro and in vivo ischemia reperfusion model. This subject researches the effect of autophagy during myocardial ischemia reperfusion pretreated by mitoKATP openers or blockers. It will find the functions and properties variation of mitoKATP during ischemia reperfusion and define the status of mitoKATP, and confirm the effect of autophagy in myocardial ischemia reperfusion pretreated by mitoKATP opener. It will clarify the protective effect of mitoKATP opener pretreatment during myocardial ischemia, and it may induce autophagy increasing through PI3K signal pathway to protect cardiac. This research will provide a new means of intervention for the prevention and treatment of myocardial ischemia and reperfusion injury.
