中文摘要: 流感相关脑病脑炎是流感的重要并发症之一,致死率高,但发病机制仍需研究。在上个国家基金资助下,我们研究结果表明流感病毒在脑内感染受限,而外周感染后会导致脑内炎症因子、氧化应激相关分子表达上调、胶质细胞激活等脑内免疫微环境变化,可能会致小鼠在学习记忆、情绪以及自身免疫状态发生改变或引发疾病,提示细胞因子风暴效应及脑内神经免疫内分泌紊乱是重要致病因素。课题提出流感病毒呼吸道感染-血浆细胞因子风暴-脑内胶质细胞激活-脑内免疫微环境变化-神经元损害-神经及精神病理效应的假想,拟系统研究流感病毒外周感染后,外周及脑部免疫微环境及神经内分泌的变化、生物学效应、及其环节和机制。明确流感病毒外周感染后所诱导的外周组织及脑部免疫微环境及神经内分泌的变化状况及时空发展规律、诱导的免疫病理损伤与效应,明确其环节和机制,完善和修正课题假想,阐明流感相关脑病和脑炎的发病机制。

英文摘要: Influenza associated encephalopathy and encephalitis is an important complication of influenza virus infection with high mortality, but the pathogenesis remains uncertain. On the base of our last NSFC project, our results showed that influenza virus infection was limited in the brain, but could cause intracerebral immune micro-environment changes, such as brain inflammation, oxidative stress related molecules expression, activation of microglia etc. In mouse model, such kinds of alter might be resulted in congnitive disorder, changes of emotion and autoimmune condition, or cause disease, suggesting that cytokine storm effect and neural-immune-endocrine disorder in the brain are important pathogenic factors. The poject put forward a hyperthesis influenza virus infection of respiratory tract - plasma cytokines in brain glial cells in the brain immune micro environment changes - neurons damage - nerve and mental pathological effects, and design to systematically study the changes of peripheral and brain immune microenvironment, the neuroendocrine changes, the biological effect, and the links and mechanism after influenza virus peripheral infection. Based on the fund, we try to revise and improve the hyperhesis, and clarify the pathogenesis of influenza related brain disease and encephalitis.
