中文摘要: 恶性肿瘤是严重威胁人类健康和生命的疾病,发展新型高灵敏度和特异性肿瘤标志物的检测方法对改善恶性肿瘤临床疗效具有十分重要的意义。本项目拟通过制备各项光学性能优异及稳定性良好的水相共轭聚合物纳米生物探针,研究共轭聚合物纳米颗粒与对肿瘤标志物有特异识别功能的抗体、基因等生物分子的相互作用和定向偶联,构建高性能(高亮信号、高稳定性、高亲和性、低交叉反应性)的共轭聚合物纳米生物复合探针;进而结合微流控技术,研究与开发用于肿瘤标志物检测的微流控生物(蛋白和基因)芯片;通过对缓冲溶液或血浆样本中多项肿瘤标志物的联合检测,评估它们在肿瘤早期诊断中的价值。希望通过结合共轭聚合物纳米生物探针和微流控芯片技术的优势,提高肿瘤标志物诊断的敏感性和特异性。同时,这一方法将共轭聚合物纳米生物探针应用于微流控芯片检测平台,对于共轭聚合物纳米信号探针在生物医学及蛋白质组学等领域的广泛应用具有重要指导意义。

英文摘要: As popular awareness of the importance of cancer early detection increases, the devices and methods which can perform quick and accurate tumor markers detection are becoming more and more compelling. Microfluidic biochip with multiple advantages,such as low sample consumption, fast response time and high throughput, appears to be a promising alternative.In this program, surface amendable conjugated polymer nanoparticles(CP NPs) with various fluorescence color will be successfully produced directly in aqueous phase by a nanoprecipitation method. The successful conjugation of biomolecules to CP NPs suggests that such NP formation has a bright future in interaction with tumor markers to construct high-quality CP bio-nanoprobe. Moreover, a novel microfluidic biochip will be developed based on CP bio-nanoprobe for simultaneous detection of multiple tumor markers, which may be useful for early cancer diagnosis. Taking advantage of the microfluidic biochip technology and the excellent fluorescence of CP bio-nanorobe, this strategy may achieve ultrasensitive and selective detection of multiple tumor markers. Also, CP bio-nanorobe based microfluidic biochip will inspire new application opportunities of CP NPs in biomedical research and proteomics.
