中文摘要: 本项目从我国重大疾病预警与诊断新方法与新技术的需求出发,以核酸适体分子识别为基础,将纳米材料和酶信号放大技术进行有机结合,建立可用于与重大疾病相关的生物标志物检测的高灵敏、高特异性荧光偏振生物传感新方法。主要内容包括:(1)纳米材料的制备、纳米探针和核酸探针的构建;(2)基于纳米材料和各种工具酶的信号放大方法建立;(3)荧光偏振适体传感器的构建及其性能研究;(4)在系统方法学研究的基础上,将所建立的新方法应用于人血清、尿液等临床样本中与重大疾病相关的生物标志物检测与验证,评估它们在重大疾病早期诊断中的应用。本项目通过结合纳米材料和酶信号放大技术,显著提高传感器的灵敏度和选择性,实现低丰度生物志物的快速检测,为重大疾病早期诊断提供技术支持,对于推动生命分析化学、生物科学和医学领域的发展具有积极的意义。

英文摘要: From the requirement of methods and technique for early diagnosis of serious diseases, this project intends to develop novel highly sensitive and specific fluorescence polarization (FP) biosensors based on aptamers, nanomaterials and enzyme signal amplification for detection of serious disease-associated biomarkers. The main content includes: (1)synthesis of nanomaterials and design of nanoprobes and nucleic acid probes; (2) development of signal amplification methods based on nanomaterials and various tool enzymes; (3) design of FP aptasensors and study on their performance; (4) the application of the developed methods for detection of serious disease-associated biomarkers in clinical samples such as human serum and urine, and evaluation of the value of these methods in early serious disease diagnosis.By combining the nanomaterials and enzyme signal amplification techniques, the sensitivity and selectivity of the proposed FP aptasensors is significantly improved, and thus the low abundance biomarkers can be detected. The developed methods will provide a new analytical technique for early diagnosis of serious diseases, and also have a positive significance for promoting the development of bioanalytical chemistry, life science and iatrology.
