中文摘要: 本课题设计以普通SD大鼠及腺苷A3受体基因转染大鼠为研究对象,观察针刺前后穴位区、非穴位区的浅筋膜形态学、腺苷A3受体、胞内第二信使cAMP、Epac、RasERK 1/2以及钙离子等信号蛋白的变化,并以原代培养大鼠浅筋膜成纤维细胞(RSF-Fibroblasts)、经 siRNA转染的RSF-Fibroblasts为对象,观察针刺对腺苷A3受体介导的 cAMP、Epac、Ras及ERK 1/2等信号蛋白的影响。从整体、细胞和基因蛋白表达层面明确浅筋膜在针刺镇痛中的重要作用。通过上调与沉默腺苷A3受体的动物及细胞模型,以及观察一系列腺苷A3受体的激动剂和拮抗剂对针刺镇痛的影响, 探讨针刺镇痛的作用机制。

英文摘要: We will use the ordinary SD rats to study on superficial fascia morphology before and after acupuncture. We will observe the change of A3 adenosine receptor, intracellular second messenger such as cAMP and calcium. We will also assess the effect of Epac, RasERK 1/2 and other signaling proteins in primary cultured rat fibroblasts (RSF-Fibroblasts). We will design, synthesize and transfect siRNA into RSF-Fibroblasts, to observe A3 adenosine receptor-mediated effects of cAMP, Epac, Ras and ERK 1/2 and other signaling proteins. We hope to explain the important role of A3 adenosine in acupuncture analgesia from whole cell gene and protein expression levels. By animal and cell models, up-regulation and silence of A3 adenosine receptors, and a series of adenosine A3 receptor agonists and antagonists , we expect to explore the mechanism of acupuncture inhibition of pain.
