中文摘要: 星形胶质细胞是中枢神经系统中数目最多的一种细胞,它作为胶质细胞的主要类别,几乎囊括了胶质细胞的所有功能。在脑缺血损伤条件下星形胶质细胞的功能调控对神经元的保护有着至关重要的影响。在先前的研究中我们发现利用δ-阿片受体激动剂(DADLE)激活δ-阿片受体(DOR),可以促进全脑缺血3天后星形胶质细胞的活化,减少了受损型星形胶质细胞,这可能是其发挥脑保护作用的一种途径。但是DOR对缺血后星形胶质细胞具体功能活动的影响还不清楚。在此基础上,本课题拟采用大鼠星形胶质细胞氧糖剥夺和全脑缺血模型,在DADLE干预下,研究DOR激活后对星形胶质细胞功能的影响及其神经元保护的PI3K/Akt信号转导机制,以期有助于寻找神经保护功能的新途径和药物作用新靶标。

英文摘要: Astrocytes are the most numerous glia cells in CNS. As major type of glia cells, astrocytes have most functions of glia cells. It is of paramount importance to neuroprotective effect by the regulation of astrocytes in the condition of ischemia. In our previous studies, we discovered that DADLE (a delta opioid receptor agonist) promoted beneficial astrocytes activation and reduced damaged astrocytes 3 days after global ischemia which may be involved in reducing their harmful effect to neurons survival. In this study, we will further investigate the morphological and functional changes of astrocytes in response to DADLE-induced DOR activation in the condition of OGD or global ischemia, and also to explore the role of PI3K/Akt signaling pathway in neuroprotection. The study will provide a new experimental evidence for finding out a new target in the treatment of brain ischemia.
