中文摘要: HCA587,又名MAGE-C2,是我室利用SEREX方法获得的一个肿瘤抗原,由于其在多种肿瘤组织高表达,而在正常组织仅局限表达于睾丸,目前被认为是制备肿瘤疫苗的理想候选分子。我们前期的研究工作显示HCA587在肿瘤细胞的表达具有促进肿瘤细胞转移的作用,且HCA587分子能与在肿瘤转移中发挥重要调控作用的STAT3(Signal Transducer and Activator of Transcription, STAT3)分子在细胞核内发生相互作用。本课题拟在前期研究工作的基础上进一步阐明HCA587对STAT3信号通路活化的影响及其作用的分子基础,并研究HCA587的表达对肿瘤细胞本身转移特性的影响,探索肿瘤抗原HCA587促进肿瘤转移的机制,为肿瘤患者提供新的预后标志物,为肿瘤治疗提供新的靶点。

英文摘要: HCA587,also known as MAGE-C2,was identified by serological analysis of recombinant cDNA expression library from heptocellular carcinoma in our laboratory. It is not expressed in normal tissues except testis, but widely expressed in a variety of malignancies. Thus it is considered as a promising candidate target for specific immunotherapy. In our previous study, we found that the expression of HCA587 in tumor cells promoted tumor metastasis in vitro and in vivo. Moreover, our study also indicated that HCA587 interacts with STAT3(Signal Transducer and Activator of Transcription, STAT3)phosphorylated on Tyr705 in the nucleus. Based on these findings, we are going to further investigate the effects of HCA587 on STAT3 signalling and the underlying molecular mechanisms,in addition , we will investigate the effects of HCA587 on the tumor intrinsic properties for metastasis to explore the mechanisms that HCA587 promotes tumor metastasis. Our study will provide a new prognostic biomarker and therapeutic target for cancer invasion and metastasis.
