中文摘要: 近来,越来越多的研究发现β肾上腺素系统和癌症的发生发展相关,但在口腔鳞癌的研究甚少。而肿瘤干细胞具有很强的自我更新和形成新的肿瘤细胞的能力,是肿瘤的起源,也是肿瘤复发、转移或化疗、放射治疗耐受的关键原因。只针对肿瘤细胞的治疗和只针对肿瘤干细胞的治疗都有其局限性,为探讨β肾上腺素受体介导的信号系统在口腔癌的表达及作用分子机制,本课题旨在研究(1)口腔鳞癌干细胞在口腔鳞癌组织中的分布和数量比例及其与口腔鳞癌发生发展相关的生物学特性(2) 各亚型β肾上腺素能受体在口腔鳞癌组织、口腔鳞癌细胞和干细胞中的表达规律(3)β肾上腺素能受体信号通路在促进口腔鳞癌发生发展中的调控作用和具体分子机制(4)β受体阻滞剂抑制口腔鳞癌发生,为β受体阻滞剂在癌症辅助治疗的临床应用提供理论基础。

英文摘要: Recently, more and more studies find out that beta adrenergic system corelate closely with the occurrence and development of solid cancers such as breast cancer and prostatic cancer, but very few studies involve in oral squamous cell carcinomas(OSCC) . Meanwhile,growing investigations show cancer stem cells have strong ability of self-renewal and proliferation,which are thought of the origin of the tumor, and key reason of tumor recurrence and metastasis or tolerance of chemotherapy and radiation therapy. Treatment target at etihter common tumor cells or cancer stem cell has its limitations.To provide a theoretical basis for the clinical application of adjuvant therapy of OSCC with beta blockers,we designed this study of the molecular mehanism of the signal system of beta adrenergic receptor mediating the deveolopment of oral cancer .The research focus on: (1) The distribution and quantity of cancer stem cells in oral squamous carcinoma tissue and their biological characteristics related to oral squamous carcinoma development (2)Distribution of each subtype of beta adrenergic receptors in oral squamous carcinoma tissues, oral squamous cancer cells and cancer stem cells (3) beta adrenergic receptors signaling pathways in promoting the development of oral squamous cell carcinomas (4)Feasibility of beta blockers suppressing morbidity and development of oral squamous cell carcinomas.
