中文摘要: 表面增强红外光谱(SEIRA)是一种高灵敏度光谱分析技术,主要以金、银等金属纳米材料为增强基底。SEIRA甚至可以实现对单分子吸附层的分析。本项目旨在研究有害细菌如大肠杆菌等的SEIRA快速检测新方法。项目拟在三维多孔材料如滤纸上固载金、银纳米粒子(NPs)构建透过式SEIRA基底,并用万古霉素等对其进行修饰以实现对微生物的主动捕获,结合物理过滤方式实现水体中大肠杆菌的捕获和富集,利用SEIRA分析的高灵敏度高特异性优点,实现对水体中大肠杆菌的定量分析。课题拟考察影响透过式SEIRA基底灵敏度的因素,如多孔材料、金属种类、NPs尺寸及形貌、固载方式等,并阐明机制。在此基础上,考察多孔材料孔径和基底上万古霉素等修饰膜对捕获有害微生物效率的影响,从而为透过式SEIRA方法用于现场有害微生物定性定量分析奠定基础。

英文摘要: Surface enhanced infrared absorption (SEIRA) is a highly sensitive spectroscopic method, which is based on substrates made of gold and/or silver nano-materials. It is reported that SEIRA is capable of detecting trace amount of analytes, even single layer of adsorbates. Here in this proposal, fast detection method for E. coli bacteria will be developed based on SEIRA technique. Firstly, we propose the immobilization of Au and/or Ag NPs of different size and shape, onto microporous 3-D materials, such as filter paper, as SEIRA substrates. Later, vancomycin and other chemicals can be used to modify the SEIRA substrates, aiming to give it active trapping capability for bacteria such as E coli. Thus, the SEIRA substrate will be able to trap the bacteria through both physical (pore) and chemical (e.g. vancomycin) effects. We will examine how the microporous 3-D materials, and the size, shape of the metallic nanomaterials, even the immobilization method for the nanomateirals, will affect the final sensitivity of the SEIRA substrates. After that, we will explore the best options for trapping bacteria on the SEIRA substrates as much as it can. We hope this strategy will lead to fast SEIRA detection method for bacteria in water.
