中文摘要: 神经递质是脑化学信使,神经递质异常与抑郁症等神经系统疾病相关。研究神经递质脑活体原位实时分析方法是神经生物化学及分析化学的前沿课题。本项目集纳米技术和碳纤维电极和超快循环伏安检测技术于一体,旨在研究生物原胺类和氨基酸类神经递质分子脑活体检测新方法。重点研究提高安培法和超快循环伏安法检测灵敏度和选择性的策略;研究纳米材料修饰电极提高检测生物原胺类神经递质灵敏度的方法;研究酶催化氧化电化学酶传感器提高监测氨基酸类神经递质灵敏度和选择性的方法;研究电极功能化提高稳定性和生物相容性的方法;研究在大鼠脑切片和脑活体中电刺激和药物刺激下神经递质监测新方法及其应用;探索检测新技术在抑郁症神经生化机制、情感和记忆过程研究中的应用。研究成果将为抑郁症等疾病的产生及诊疗研究提供科学方法手段,将促进脑生理学、病理学及药理学的研究,为从分子水平上开展情感与记忆过程研究提供关键方法和技术。

英文摘要: Neurotransmitters are the brain’s chemical messengers and the neurotransmitter abnormalities are associated with depression nervous system diseases. The aim of this project is to develop novel electrochemical methods in vivo for the detection and quantification of brain neurotransmitters including the biological amines and amino acid neurotransmitters with functional carbon fiber microelectrodes in vivo incorporating nanotechnology and ultrafast cyclic voltammetry. The project will be focused on strategy to improve the sensitivity and selectivity of the amperometric and ultrafast cyclic voltammetric methods for brain neurotransmitters in vivo. The main research work includes: to study nanomaterials modified electrode to improve sensitivity of the methods for the detection of biological amine neurotransmitters; to design and fabricate the enzyme-catalyzed oxidation electrochemical biosensors to improve sensitivity and selectivity of the methods for monitoring of amino acids nerve neurotransmitter; to design and fabricate functional working electrodes and reference electrodes to improve potential stability and biocompatibility of the electrodes; to study applications of the quantification monitoring methods developed in cultured rat brain slices and in living rate brains under the electrical and drug stimulation
