中文摘要: 玉米醇溶蛋白是玉米籽粒中最主要的储藏蛋白,直接影响玉米籽粒的蛋白品质。玉米醇溶蛋白种类繁多,并且调控复杂。然而目前已经克隆的转录因子仅2个(分别为Opaque2和PBF),并不能覆盖所有主要类型的醇溶蛋白基因的转录调控,其中包括玉米籽粒中含量最高的19kD醇溶蛋白基因。我们前期通过对19kD醇溶蛋白基因核心启动子的酵母单杂交筛选等研究,获得了两个新的转录因子,分别专一结合19kD醇溶蛋白基因核心启动子区,以及位于核心启动子区上游的关键顺式元件P-box,分属于锌指类和bZIP类转录因子。本研究将在此基础上,进一步研究这两个转录因子的表达特征和对靶标基因的转录调控作用;通过蛋白互作等研究探讨它们可能的调控网络;并通过转基因过表达和抑制表达等手段,探索这两个基因的生物学效应和可能的应用前景。本研究将显著推进我们对醇溶蛋白基因转录调控的认识,为利用和改造玉米储藏蛋白奠定理论基础。

英文摘要: Zein proteins are the most abundant storage proteins in maize seeds, and a key determinant for maize seed protein quality. Zein proteins are encoded by many different genes, with complicated regulations. However, so far only two transcription factors (Opaque2 and PBF, respectively) were cloned. They certainly could not explain the transcription regulation for all major zein protein classes, in particular for the most abundant 19kD zeins. In our previous study, we identified two new transcription factors by yeast one hybrid screening, using 19 kD zein gene core promoter and the Prolamin box as screening baits. They belong to zinc finger type and bZIP type transcription factors, respectively. In this study, we will characterize their features as transcription factor genes, and their transcription regulation to the target genes. We will explore their potential regulation network using protein-protein interaction analysis. We will evaluate their biological effects and potential application value by over expression and gene suppression with transgenic approach. The results from this study will extend our knowledge about the transcriptional regulation of zein genes, and lay the foundation for further genetic modification of storage proteins in maize.
