中文摘要: 有机分子光开关螺吡喃和螺噁嗪的研究主要集中在它们的光致变色效应及其在分子电子学与光学器件中的应用,而有关螺吡喃和螺噁嗪基团自身的荧光及其可逆性研究报道较少。本项目首先利用乳液聚合,将螺吡喃和螺噁嗪等光开关单体与常用乙烯基单体共聚合得到具有可逆荧光的聚合物纳米颗粒,研究纳米颗粒在溶液和薄膜中的光致变色和光致发光效应;通过单分子显微镜研究单个纳米颗粒的可逆荧光效应;将纳米颗粒与生物分子偶联,利用纳米颗粒的可逆荧光信号进行DNA及蛋白质检测;将纳米颗粒与靶向细胞表面受体的抗体分子偶联,用于细胞的荧光成像。预计本项目将通过改变螺吡喃和螺噁嗪等光开关分子的微观分子环境以提高其荧光量子效率和可逆性能,加深对该纳米颗粒可逆荧光效应的理解,开创基于螺吡喃和螺噁嗪等光开关分子可逆荧光的应用,特别是为生物医学研究提供一种新型的可逆荧光探针,有利于降低生物分子传感及细胞成像研究中假阳性结果和细胞自荧光的干扰。

英文摘要: There are hundreds of reports about organic molecular photoswitches including spiropyrans and spirooxazines every year, which have been focused on their photochromism and application in molecular electronics and photonics. However, the research on fluorescence and its reversibility is scarce. Here we first utilize emulsion polymerization to synthesize the copolymer nanoparticles containing photoswitchable monomers affording polymer nanoparticles with reversible fluorescence, investigate the photochromism and photoswitchable fluorescence of the resulting polymer nanoparticles in solution and thin film, use single molecular microscopy to focus on the reversible fluorescence of single nanoparticle, and prepare nanoparticle bioconjugates for the detection of DNA and proteins as well as cellular fluorescent imaging. It is anticipated that the fluorescence quantum yield and reversibility of nanoparticles as well as reversible fluorescence effects will be advanced through changing the microscale environment of molecular switches in nanoparticles. We are going to enhance the understanding and application of the reversible fluorescence effects, especially provide a reversible fluorescence probe to lower the interference of the false results and cellular self-fluorescence in biomolecular sensing and cellular imaging.
