中文摘要: 猪流行性腹泻病毒(PEDV)感染的爆发,给养猪业带来严重危害。病毒与宿主细胞互作机制是抗病毒研究的理论基础。目前,在宿主细胞中,PEDV感染相关蛋白尚未得到深入研究。整合素在多种病毒感染中发挥重要作用,日趋成为研究热点。我们前期研究发现,整合素β3与PEDV感染具有相关性。为了进一步明确整合素β3在PEDV感染过程中的作用,本研究拟通过靶蛋白表达细胞系构建、靶基因沉默与过表达、流式细胞术、激光共聚焦显微镜、免疫共沉淀和GST-pull down等方法和技术,在PEDV非敏感细胞系和易感细胞系中,明确整合素β3与PEDV的结合作用,介导PEDV的侵入作用,与PEDV 纤突蛋白(S)的相互作用,以及与PEDV受体猪氨基肽酶N(pAPN)的协同作用,为PEDV感染机制理解及抗病毒药物设计提供理论基础。

英文摘要: The outbreak of porcine epidemic diarrhea virus (PEDV) infection has led to severe economic losses in the pig industry. Interaction mechanism of virus and host cells is important theoretical basis of antiviral research. At present, PEDV infection-associated proteins in host cells are not clearly understood. Integrin plays an important role in a variety of virus infections, and is becoming a research focus. In our preliminary study, integrin β3 showed an association with PEDV infection. To confirm the role of integrin β3 in the process of PEDV infection, the techniques and methods,involving in construction of the target protein-expressed cell lines, over expression and silencing of the target genes, flow cytometry, laser scanning confocal microscopy,Co-IP, and GST-pull down, were used for analysis of binding of PEDV with integrin β3, integrin β3-mediated virus entry, interaction of PEDV S protein with integrin β3, and collaboration of integrin β3 with pig aminopeptidase N (pAPN) in sensitive and unsensitive cell lines of PEDV. Theses data will increase the understanding of PEDV infection, and provide some basis for antiviral drug designing of PEDV.
