中文摘要: 猪流行性腹泻病毒(PEDV)引起的仔猪腹泻是近年来严重危害我国养猪业的一种重要传染病,现已证实新流行的PEDV基因组发生明显变异,但目前对于PEDV感染致病的机理尚不明晰,病毒侵染细胞是病毒发挥其致病能力的关键环节,揭示病毒与宿主细胞之间的相互作用是阐明PEDV感染致病机制的基础,本项目以PEDV新流行毒为实验材料,拟从解析纤突(S)蛋白在介导PEDV侵染细胞过程中的分子细节入手,利用BiFC系统、CO-IP、质谱分析等病毒分子生物学试验技术和方法,从病毒与感染细胞两个方面,探索PEDV的S蛋白与其受体蛋白互作的功能域及其作用途径,挖掘Vero细胞中与PEDV编码蛋白相互作用的未知受体蛋白,分析其对PEDV感染、复制与增殖能力的影响,探讨PEDV S蛋白在病毒侵染细胞过程中的生物学意义,为阐明PEDV感染和致病机理以及抗PEDV感染有效药物的研制提供理论依据。

英文摘要: PEDV has received an increasing attention as the important pathogen causing diarrhea in piglets. Studies has confirmed the genomic mutation in recent strains. However, less is know about the pathogenesis of PEDV. Intracellular infection of virus is the key step for virulence. Exploration of the interaction between PEDV and host cells is necessary for understanding the pathogenesis of PEDV. The aim of this study is to investigate the receptor of S protein of PEDV for cell infection, and screen the other protein-protein interaction of PEDV proteins in Vero cells, by using BiFC, CO-IP and mass spectrum. It is expected to clarify the mechanism of S protein in intracellular infection, and to support the future studies of PEDV pathogenesis and medicine development against PEDV infection.
