中文摘要: 猪流感是影响养猪业的重要疫病之一,具有重要的公共卫生意义。探讨猪流感病毒感染后宿主的免疫反应对该病的防控意义重大。为阐明线粒体抗病毒信号(MAVS)蛋白这一新型免疫分子在抗猪流感病毒感染的先天性免疫中的作用,本项目拟用H3N2亚型猪流感病毒感染宿主原代细胞(巨噬细胞、树突状细胞、气管上皮细胞),检测与抗病毒先天性免疫相关的两条重要信号通路上的相关受体(TLRs、RIG-I/MDA5)、重要接头蛋白(MAVS、MyD88、TBK1、IRF3/7、NF-κB等)及效应分子(促炎症细胞因子及Ⅰ型干扰素),分析其活性变化规律;进而用TLR7/8拮抗剂及靶向MAVS的特异性siRNA,观察TLR7/8抑制和MAVS信号通路阻断对宿主抗病毒反应的影响;最后通过动物试验,全面揭示MAVS蛋白在猪流感病毒与宿主互作中的作用。研究有助于深入了解宿主识别和清除病毒的免疫机制,为流感病毒的防控提供新的思路。

英文摘要: Swine influenza is common and economically important in pig populations worldwide. It is also a paramount public health concern. Understanding the immune response of the host is requisite in controlling infection. Recently, mitochondrial antiviral signaling (MAVS) protein was demonstrated to have antiviral activity. To evaluate the antiviral role of MAVS on H3N2 swine influenza virus infection in vitro, receptors (TLR7/8, RIG-I/MDA5), adaptor proteins (MAVS, MyD88, TBK1, IRF3/7, NF-κB) and effector factors (inflammatory factor and interferon-I) are quantified. Furthermore, TLR7/8 antagonist and specific siRNA targeting MAVS are utilized to evaluate the effect of the inhibited TLR7/8 and MAVS on the ability of the host to develop immunity to the virus. Finally, animal experiments are done to demonstrate the antiviral role of MAVS in virus-host interactions. This study will contribute to thoroughly understanding the mechanism of immune recognition and clearance of viral infection of the host, and may offer new thoughts to the prevention and control of influenza virus infection.
