中文摘要: 蛋白质吸附是生物医学、生物材料和生物工程等交叉学科的关键科学问题。其中,蛋白质吸附过程中的构象动态变化,一直是实验检测的瓶颈所在。本项目结合手性和频光谱解析界面蛋白质结构的独特能力和光外差探测技术增幅相敏的优势,发展新型的手性相敏和频光谱(Chiral-PS-SFG),从而实现原位实时测量界面蛋白质取向和结构。选择牛血清白蛋白(BSA)和牛纤维蛋白原(BFg)为模型蛋白质,通过改变外界因素(温度、PH值、离子浓度等),表面特性(镀高分子膜、镀金属膜),乃至蛋白质本身的性质(结构域修饰),观察蛋白质在熔融石英玻璃表面上的取向和结构的动态变化及规律,在分子层面上考察蛋白质结构-吸附特性-相互作用之间的关系,以期能够理解、控制并利用或阻抗蛋白质在界面上的吸附。该研究不仅为动态检测蛋白质吸附的构象变化提供了一种新的思路和方法,而且可以为设计生物相容材料和生物传感器等提供实验基础和理论依据。

英文摘要: Protein adsorption is a crucial process in many cross-disciplinary areas such as bio-medicine, bio-materials and bio-engineering. However, monitoring conformational changes of proteins at interfaces has long been a bottleneck in protein adsorption process. This project aims to observe in situ and in real-time the protein orientations and structures at interfaces by developing the chiral phase-sensitive sum frequency generation (Chiral-PS-SFG), for which we take the advantage of the Chiral SFG in probing protein structures at interfaces and the Heterodyne-Detected SFG for signal amplification and phase sensitivity. Using bovine serum albumin (BSA) and bovine fibrinogen (BFG) as the model proteins, protein adsorption onto silica surface will be studied. By changing solution properties (temperature, pH, ion concentration, etc.), surface properties (modified by polymer film or metallized film), and even protein properties (modified at protein domain), the conformational changes and dynamics of adsorbed proteins at interfaces will be probed directly.
