中文摘要: Akt信号通路与非小细胞肺癌(NSCLC)的发生发展密切相关。我们在NSCLC组织中初步筛选出差异表达的Akt信号通路相关蛋白,发现AKT2不仅在NSCLC组织中高表达,而且是肺癌患者不良预后的重要影响因素;沉默Akt2基因,能够明显抑制肺癌细胞的增殖,侵袭及细胞成瘤能力。因此,我们推测Akt2可能是Akt信号通路的核心蛋白,在NSCLC发生发展中起到关键作用。但目前Akt2的分子作用机制尚不明确,本项目在前期研究基础上,进一步阐明Akt2在NSCLC中发挥作用的关键环节,通过RNA干扰及构建过表达载体,结合体外细胞模型、动物模型及临床标本的研究,明确Akt2的下游靶基因,探讨AKT2 介导的信号调控机制,为肺癌基础研究提供新的理论依据,为肺癌靶向治疗提出新的靶点。

英文摘要: Akt signaling pathway has been reported to contribute to tumorigenesis in Non-small cell Lung cancer.In our research, we analyzied the expression profile of Akt singnaling related molecules. Our results showed that Akt2 expression was higher in NSCLC compared with it in normal lung tissue, and Akt2 expression level was correlated with poor prognosis in NSCLC.Knockdown of Akt2 in A549 cells significantly inhibited cell proliferation, invasion and tumor growth. Therefore, Akt2 may be a central molecule in Akt signaling pathway in NSCLC. However,the molecular mechanism is still unclear. In this project, based on previous results, we explore the role of Akt2 and the underlying molecular mechanism in vivo and vitro.
