中文摘要: 骨质疏松症是一种以骨密度(BMD)降低、骨微结构破坏和脆性骨折发生风险增加为主要特征的慢性复杂性疾病,中医药防治骨质疏松症具有较大的潜力与优势。肾虚血瘀是导致OP的主要病理机制。依据其病机制定的补肾活血方能够增加OP患者的骨矿含量、 提高骨密度,其作用机制需深入研究。本研究首先建立去卵巢大鼠骨质疏松动物模型,观察补肾活血方对去卵巢雌性大鼠骨密度及骨代谢指标的影响,及大鼠骨形成Ⅰ型胶原、相关蛋白MBP2及Hedgehog信号通路相关SHH/IHH蛋白表达的作用机制,PCR芯片检测Hedgehog通道, RT-PCR、Western Blotting进行验证。体外观察补肾活血方对SD大鼠中骨髓基质细胞的成骨能力的影响,观察碱性磷酸酶活性、茜素红染色及Ⅰ型胶原、MBP2蛋白的表达及RT-PCR、Wstern Blotting验证,进一步探讨补肾活血方治疗绝经后骨质疏松症的作用机制。

英文摘要: Osteoporosis is a kind of chronic complex disease which the main characteristics occur as bone mineral density (BMD) reduce, bone microstructure damage and increased risk of brittle fracture, Traditional Chinese medicine has great potential and advantages in preventing and treating osteoporosis. Renal deficiency and blood stasis is the main pathological mechanism which could cause OP. Based on this pathological mechanism, Bushen Huoxue Recipe is made to Increase OP patients' bone mineral content and to Improve patients' bone mineral density; the mechanism of action should be further research. This study first establish the ovariectomized rats animal model with osteoporosis, to observe the influence of the Bushen Huoxue Recipe to ovariectomized female rats' bone mineral density and bone metabolic marker, and form the type I collagen、related protein MBP2 and the mechanism of action of Hedgehog signaling pathway related expression of SHH/IHH protein,PCR chip inspection Hedgehog signaling pathway, RT-PCR、validate the Western Blotting. to observe the influence of Bushen Huoxue Recipe to the osteogenic capability of bone marrow stromal cells of SD rats in vitro, to observe the alkaline phosphatase activities、staining of alizarin red and type I collagen、expression of MBP2 protein and confirm RT-PCR、Wstern Blotting, to explore further the mechanism of action of Bushen Huoxue Recipe to treat the postmenopausal osteoporosis.
