中文摘要: 核苷肽类抗生素(peptidyl nucleoside antibiotics)是一类由核苷和多肽经过系列的酶促生化反应形成的有生物活性的次级代谢产物,其中核苷和多肽是如何缩合的,一直是国际上非常关注,但至今尚未被揭示的前沿科学问题。本项目以尼可霉素、多氧霉素和谷氏菌素为研究对象,对该类抗生素的核苷和肽基缩合机制进行深入研究。拟采用遗传学和生物化学等方法,研究与缩合相关的基因的功能,酶学生化特性及其催化机制,揭示核苷肽类抗生素缩合的分子机制,为突变生物合成、组合成生物合成等手段得到新型的核苷肽类抗生素提供重要的依据。

英文摘要: Peptidyl nucleoside antibiotics are a group of secondary metabolites consisting of nucleoside and peptidyl moieties.They display a broad spectrum of biological activities, such as antibacterial,antifungal,antitumor,antivirus, etc. This project aims to reveal the mechanism of condensation between peptidyl and nucleoside moieties of nikkomycin, polyoxin and gougerotin. For this purpose, we will investigate the functions of genes involved in the assembly process. In addition, the enzymatic properties of their coressponding enzymes will also be studied extensively.These studies will be useful for the generation of novel peptidyl nucleoside antibiotic derivatives by mutasynthesis and combinatorial biosynthesis.
