中文摘要: 机体衰老时伴随着机体内氧自由基的生成增多和清除能力减低,氧自由基的过量积累会损伤血管内皮细胞的功能,加速血管老化并诱发内皮细胞功能障碍相关的疾病。申请者前期在细胞水平的研究发现,硫化氢(H2S)能通过抗氧化应激作用延缓培养的人脐静脉内皮细胞的老化进程。本项目拟在原有研究的基础上,以H2S调控内皮细胞氧自由基水平为切入点,从整体水平和细胞水平深入研究H2S延缓内皮细胞老化的机制,并从内皮细胞NO的合成和释放、NO合成酶eNOS脱偶联水平和以NO为基础的蛋白质巯基亚硝基化三个方面的变化进行阐述。本研究将明确H2S是否能有效的保护血管内皮细胞功能、减少内皮细胞功能障碍的发生,从而延缓血管老化过程,并为血管老化相关疾病的临床治疗提供新的治疗策略和思路。

英文摘要: Senescence usually accompanied with the increased formation and decreased removal rate of oxygen free radical, the abundant accumulation of oxygen free radicals will damage the endothelial cells and lead to the diseases which related to vascular endothelial dysfunction. In prior study, the applicant found that the third gas molecular, hydrogen sulfide (H2S), can postpone the aging process by anti-oxidative stress in human umbilical vein endothelial cells. The role will be further studied in this project. We will explore the specific mechanisms of H2S against the aging process of endothelial cells including changes of synthesis and release of endothelial NO, the uncoupling of eNOS (endothelial NO synthase) and NO-based protein S-nitrosylation in overall level and cell level. This study will identify the effective protection of H2S on vascular endothelial cells from endothelial dysfunction and the aging process of blood vessels, and supply new therapeutic strategies and ideas on clinical treatments of diseases which related to aging process of blood vessels.
