中文摘要: 嗜热酶热稳定性的分子机制研究,不仅是食品酶学领域中的研究热点,也是提高食品酶耐热性及扩大其应用范围的关键,因此开展嗜热甘露聚糖酶热稳定性分子机制的研究具有重要的理论和现实意义。申请人前期在嗜热甘露聚糖酶热稳定性的研究中发现,除糖基化修饰和盐桥对酶的热稳定性有较大影响外,Loop也能显著影响酶的热稳定性,而其内在的分子机制还有待阐明。本项目以嗜热甘露聚糖酶Loop为研究对象,利用现代生物信息学技术,分析嗜热甘露聚糖酶Loop的分子特征;结合同源建模、分子动力学模拟、定点突变和圆二色谱分析等技术,揭示Loop影响嗜热甘露聚糖酶热稳定性的分子机制;依据上述研究结果,探讨理性改造Loop提高酶热稳定性的技术方法。本项目的研究不仅仅可以丰富嗜热酶的热稳定理论,也可为寻求新的提高酶热稳定性的策略提供一定的理论指导。

英文摘要: The study of the thermostability of thermophilic enzyme, not only is the hotspot of protein engineering research, but also is the key of improve the heat resistance and expand the range of application. Therefore, it is a very important that research on the molecular mechanism of the thermal stability of thermophilic enzyme. We have obtained four thermophilic mannanase from the thermophilic microorganisms. In addition to N-glycosylation modification and salt bridg influence the thermal stability of thermophilic enzyme, we found that Loop is another important factor in the thermal stability of the enzyme. However, It has not been knew that the role of Loop in thermal stability of the thermophilic mannanase. In this study, we use the bioinformatics analysis the characteristics of Loop, and use Homology Modeling technology, Molecular dynamics simulation technology, Site-directed mutagenesis technology and Heterologous expression technology, reveals the molecular mechanisms of loop affect on the thermal stability. This research reveal on the molecular mechanism of mannanase enzyme thermal stability. Not only that, but also provide a new strategy of improving thermal stability of mannanase.
