中文摘要: 处在光合作用起点的光系统II是光合膜上最重要的蛋白复合体,催化光驱动的水裂解放氧反应并介导原初电荷分离和电子传递。过去的研究使人们对PSII的主要组成和结构有所了解,但对该复合体特别是超级复合体形成和/或稳定的分子机理认识还很有限。因此,本项目拟围绕这一重要科学问题,以光合模式生物-莱茵衣藻为材料,从已筛选的光合突变体X32入手,在基因和蛋白水平以及对光逆境等响应等方面进行深入研究,揭示该基因及其编码蛋白在衣藻PSII生物发生及功能调控中的作用。首先,采用已优化的衣藻基因克隆方法,获得靶基因的全长序列。其次,运用生物信息学软件对其进行序列分析和功能预测。进一步采用系统生物学方法和光合生理生化方法, 揭示该基因及其编码蛋白在衣藻光合系统II生物发生及光保护中的重要作用。

英文摘要: Photosystem II is the initial and most crucial multisubunit protein complex catalizing the light-driven reactions of water-spliting and primary charge separation and electron transfer. Though the composition and structure of PSII have been understood in great deal, our knowledge on the molecular events of its biogenesis and regulation mechanisms is still limited, especialy regarding the formation and maintenance of the PSII supercomplex. In this research project, a mutant strain (x32) of Chlamydomonas reinhardtii, which exhibits impaired photosynthetic PSI phenotype, will be used throughout the investigations. Firstly, the mutated gene will be isolated by molecular cloning. Secondly, the functional significance will be examined using systems biological means and biochemical analysis. The experimental data to be obtained will bring about novel insights regarding PSII biogenesis and photoprotection as well as valuble gene(s) toward enhanced efficiency of photosynthesis in Chlamydomonas.
