中文摘要: 脑胶质瘤是颅内发病率最高的恶性肿瘤,传统的治疗方法效果十分有限。HER2是在多种人肿瘤中高表达、且在肿瘤的发生和转移中发挥着重要作用的表皮生长因子受体家族分子。HER2在正常人的中枢神经系统中不表达,但是在脑胶质瘤中,胶质瘤细胞表面的HER2过表达与其恶性表型密切相关,因此它可以成为人胶质瘤治疗的特异性靶点。我们在研究中探讨了四种人胶质瘤细胞系A172、U251、U87和SHG-44中HER2的表达情况。通过将HER2单链抗体与白喉毒素转膜结构域及促凋亡分子活性caspase6的融合,构建出能识别HER2阳性的肿瘤细胞,并转位入细胞内杀伤瘤细胞的靶向促凋亡蛋白。在试验中验证靶向融合蛋白对HER2阳性胶质瘤的杀伤效果,从而为恶性胶质瘤的靶向基因治疗提供了新的途径。

英文摘要: Glioma is the most common tumor of the brain. Though the aggressive surgery or radiotherapy, patient prognosis still poor. HER2 is over expressing in several human tumors. When it is active, HER2 promote the ability of tumor cells in metastasis and invasion. HER2 is negative in the human central nervous system. The over-expressing of HER2 is associated with malignant phenotypes of human gliomas. HER2 can be a target for targeted therapy of gliomas. We investigated the expression of HER2 in glioma cell lines A172, U251, U87 and SHG-44. We constructed a novel immune caspase6 which were fused with e23sFv, Fdt and active caspase6. The novel immune caspase6 can bind to and be internalized into HER2 overexpressed tumor cells, then cleaved by furin and translocated into the cytoplasm directly. Our investigation provides a novel strategy for the treatment of HER2 overexpressed gliomas.
