中文摘要: 西瓜生产中存在严重裂果的问题,由于其裂果的遗传机制和分子机理研究非常有限,善无有效解决办法。本项目在研究西瓜裂果性状的主基因+多基因遗传效应和遗传模型基础上,利用RIL群体构建分子标记连锁图谱,分析不同环境条件下西瓜裂果性状QTL的遗传及其与环境互作效应;利用NIL群体,通过SSH技术构建抗裂和感裂相关基因特异表达的cDNA文库,获高丰度的抗感裂果相关差异表达的基因或EST 序列;通过对相关候选基因表达验证,发掘西瓜抗裂果相关基因。为解析西瓜裂果机制及抗裂新品种选育技术奠定基础。

英文摘要: Fruit cracking is a serious problem in watermelon production, and it still has no effective solution because of limited study on the genetic mechanisms and molecular mechanisms of fruit cracking. We studied the inheritance mode of fruit cracking by applying the joint segregation analysis on the mixed genetic model of major gene plus polygene in six generations of watermelon in the past years. In this project, the population of recombinant inbred lines (RIL) derived from the cross of fruit cracking susceptible inbred line L-103 and fruit cracking resistance germplasm X-101 will be used to construct the molecular linkage map to analyze the locations of fruit cracking trait QTL and environmental interaction effects in different environmental conditions. The population of near isogenic lines (NIL) derived from the foreground selection for the major QTL and the background selection for the genome of the recurrent parents will be used to construct the cDNA library of fruit cracking related genes, obtaining high abundances of differentially expressed fruit cracking resistant and fruit cracking susceptible genes or EST sequences. The candidate genes were verified by RT-PCR to explore the fruit cracking resistant related genes in watermelon. The results will lay the foundation for the analysis on the fruit cracking mech
