中文摘要: 多模态成像技术是多种成像模式的结合与优势互补,是恶性肿瘤早期诊断的有力手段。适配体是体外人工合成的ssDNA 或RNA 寡核苷酸分子,为一种潜在理想的活体成像探针。Endoglin 是一种细胞膜糖蛋白,与肿瘤血管生成密切相关,是一种新型的肿瘤分子靶点。本课题拟利用前期研究筛选的Endoglin适配体为靶标识别分子,以磁性壳聚糖纳米粒为载体,Cy5.5为近红外荧光标记分子,构建针对肿瘤新生血管内皮细胞的Endoglin有特异性识别能力的荧光/磁性Endoglin适配体纳米靶向探针。通过与鼠肺癌新生血管内皮细胞的体外实验,验证该探针的稳定性、靶向性及生物学特性。然后,以荷人肺癌裸小鼠为肿瘤动物模型,研究探针在鼠体内的MRI/NIRFI双模态活体靶向成像。为适配体多模态靶向分子探针尽早用于临床奠定理论基础和提供实验依据。

英文摘要: Multimodal imaging is one of the effective means for early diagnoses cancer due to the combination of the advantages of molecular imaging techniques. Aptamer, short single-stranded DNA or RNA oligonucleotides, is a promising molecular probes in vivo cancer imaging. Endoglin, a cell surface molecule over-expressed on endothelial cells of tumor-associated blood vessels, is a new type of tumor molecular targets site. In this project, a multifunctional of fluorescence/magnetic Endoglin aptamer targeting molecular nanoprobe is prepared with magnetic chitosan nanoparticles as multifunctional nanometerial carrier, Cy5.5 as near infrared fluorescent material, and Endoglin aptamer as target recognition group. The aptamer imaging nanoprobes' targeting as well as the target cell biological effects is evaluated. At the same time, the feasibility, validity and mechanism of the near-infrared fluorescence imaging (NIRFI) and magnetic resonance imaging (MRI) are discussed in vivo and in vitro using lung cancer as the research object. The application of aptamer nanoprobes targeted to Endoglin might hold great promise in the detection of tumor living cells and in vivo cancer imaging, and lay the theoretical foundation in clinical diagnosis.
