中文摘要: 由于稀土配合物具有特殊的荧光性质,多年来一直被生物医学荧光成像领域所关注。本项目将利用稀土配合物的荧光特性,结合生物医学荧光成像探针的设计思路,研究和开发一类同时具有荧光成像,跨膜和靶点识别特性的生物荧光成像分子,为其能在临床检测中得到应用打下理论基础。 本研究中,将具有良好跨膜性质的多肽(tat)选为目标分子的主链,在多肽的侧链上接入,可以选择性识别线粒体的三苯基膦和以多足体配体配位的稀土配合物。固相多肽合成和选择性氨基保护是目标分子合成的主要技术。本课题首先探索和优化目标分子的合成路线,表征方法及荧光性质;然后,结合细胞实验,探讨目标分子的毒性,生物相容性以及在生物荧光成像中的应用;最后,通过对此方法和现有方法的总结和对比,得出自身方法的优缺点,使之在今后的生物靶点成像的研究中得到改进和发展。

英文摘要: The unique luminescence properties of lanthanide chelate complexes have been proved their value in the field of biomedical fluorescence imaging in many years. Base on the design principles of biomedical fluorescence imaging agents, a new kind of molecules equipped with rare earth complexes luminescence characteristics may be designed and synthesized with the multi-functions, such as biomedical fluorescence imaging, transmembrane, and target identification. Finally, this type of molecules will be applied to clinical research. In this study, the transmembrane peptide (tat) will be chosen as the main chain of the target molecules. On the side chain, triphenylphosphine ligand will be used as a ligand which can selectively identify mitochondrial and lanthanide chelate complexes with the podands ligand will be selected as the biomedical fluorescence imaging agent. The solid phase peptide synthesis and selective amino protection strategy will be the main technique tools to synthesize target molecules. Firstly, this proposal will explore and optimize the synthetic route of target molecules, study characterization methods, and measure their fluorescence properties; secondly, the toxicity, biocompatibility, and bioluminescence imaging of the target molecules will be done by cell based assays; Finally, comparing the comm
