中文摘要: 缺氧是大多数实体肿瘤的标志之一,和肿瘤的治疗与预后直接相关,而临床缺乏直接的缺氧造影。本项目利用铱配合物的磷光发射对氧敏感的特点,提出发展含铱配合物的肿瘤缺氧敏感的近红外高分子探针,对肿瘤的缺氧进行活体成像检测。针对小分子铱配合物的非水溶,可见光发射,血循环时间短的问题,通过对铱配合物主配体的结构调控和对辅配体的聚乙烯基吡咯烷酮化,使探针具有近红外发射,水溶性与生物相容性好,肿瘤缺氧敏感性高,血液循环时间长,肿瘤富集程度高,体内分布合理的特点。进一步地通过在配体中引入其他肿瘤环境敏感的基团构建多功能高分子探针。在细胞和动物二个层面上深入考察这些探针分子对缺氧的敏感响应,对肿瘤(包括植入和转移瘤)的诊断效果,探索探针对肿瘤化疗效果的跟踪,揭示探针结构与缺氧响应的规律,关联缺氧与肿瘤预后及耐药的关系,为肿瘤的诊断(包括早期诊断)提供可靠的探针和方法。

英文摘要: Hypoxia is a feature of most solid tumors, and associates with treatment outcome and prognosis. However, hypoxia imaging in clinic remains great challenges. Here the cancer hypoxia-sensitive and near infrared emitting polymer probes containing Ir(III) complex moiety are proposed to image cancer hypoxia in vivo by taking advantage of oxygen sensitive phosphorescence quench of Ir(III) complexes. In contrast of lipophilicity, visible emitting and rapid clearance of small molecular Ir(III) complexes, the designed polymer probes are near infrared emitting, water-soluble, biocompatible and long circulating after the ligands are chemically modified with large conjugated structures and water-soluble poly(N-vinylpyrrolidone). We will evaluate the hypoxia imaging performance of the probes in cells and tumor-bearing mice, and correlate the probe structures with hypoxia imaging in vivo. It is expected to establish the relationship between tumor hypoxia and prognosis or drug-resistance. By this project, we hope to provide the sensitive and robust probes for cancer diagnosis.
