中文摘要: 石斛多糖是我国传统药材石斛的主要活性成分,我们的预实验证实其具有防治肝纤维化的作用,但具体机制尚不明确。近年来肠道稳态的破坏被证实促进肝纤维化的形成,同时植物多糖对于肠道稳态的调控正受到广泛的关注。本课题以肠道稳态为切入点,建立硫代乙酰胺破坏肠道稳态诱导肝纤维化的模型,探索石斛多糖通过调节肠道菌群和保护肠道黏膜屏障,进一步防治肝纤维化的作用及机制。体外应用正常肠上皮细胞IEC-6和Caco-2细胞模拟的肠黏膜屏障模型,深入阐释石斛多糖抑制肠上皮细胞凋亡,保护细胞间的紧密连接,从而恢复肠道稳态作用的分子机制。通过本课题的研究,有望为肝纤维化的治疗开创了全新的研究视角和作用靶点,同时为寻找天然高效防治肝纤维化的活性成分提供新的思路和途径。

英文摘要: Polysaccharides isolated from dendrobium have been confirmed the preventive effects on hepatic fibrosis in our previous study, but the mechanism is remained to further investigation. Recent reports have demonstrated that the disruption of gut homeostasis is associated with the progression of hepatic fibrosis, and it is widely concerned that plant polysaccharides may play an important part in regulating gut homeostasis. This study gets the gut homeostasis as the entry point and establishes a model of hepatic fibrosis, in which thioacetamide causes destruction of gut homeostasis. Based on the model, the regulation effects of dendrobium polysaccharides for gut flora and gut mucosal barrier are to be investigated and the further exploration of related mechanisms can also be conducted. In vitro the protective role of dendrobium polysaccharides was assessed by incubating IEC-6 and Caco-2 cells. We have further investigate the molecular mechanisms of dendrobium polysaccharides inhibited the apoptosis of IEC-6 and improved the barrier dysfunction of Caco-2 cells. This research is expected to create a brand new perspective and targets for the treatment of hepatic fibrosis. And modulation of the gut homeostasis may represent a novel anti-fibrotic therapeutic approach.
