中文摘要: 树鼩因与灵长类最近的亲缘关系和诸多独特的特点,在生物医学研究中被认为是最有望于替代珍贵的灵长类动物之新型实验动物。现有文献报道了相当多的树鼩基础生物学数据,但是树鼩精子获能、顶体反应及其分子机制方面的研究,迄今尚未见有PubMed 收录论文发表,这势必会成为制约树鼩在疾病机理研究和新药研发中广泛应用的瓶颈。我们的前期工作表明树鼩精子获能伴随有蛋白质酪氨酸磷酸化,此过程为时间、BSA、Ca2+、HCO3-依赖性的cAMP-PKA信号转导途径,研究结果为进一步研究树鼩精子获能的分子机制奠定了良好的基础;我们还发现树鼩成熟精子中有SRC和ADF/cofilin蛋白表达,因此,我们提出SRC、ADF/cofilin、PLCγ以及PIP2相互作用,完成对树鼩精子顶体反应的调控之假说。本项目能够促进树鼩生殖与发育生物学的发展,具有重要的理论意义和现实意义。

英文摘要: Due to its genetically close relationship to primates and many other unique characteristics, the tree threw has been proposed as an alternative experimental animal to precious primates in biomedical research. Quite a few fundamental biological data on tree threws have been published. However, no research on sperm capacitation, acrosome reaction (AR) and their molecular mechanisms for this animal has been reported in PubMed. This situation inevitably will become the bottle neck that restricts tree threws' wide use in disease mechanism and new drug development. Our preliminary study demonstrates that sperm capacitation of tree threw is accompanied by tyrosine phosphorylation in a subset of proteins, and this process is a cAMP-PKA dependent signaling pathway that is reliant on time period, BSA, Ca2+, HCO3- during sperm capacitation. The result provides a good basis for our further understanding the molecular mechanism of sperm capacitation in tree threws. In addition,we find proteins of SRC and ADF/cofilin in the mature tree threw sperm. Thus, we hypothesize that the interreaction among SRC, ADF/cofilin, PLCγ and PIP2 is responsible for AR regulation in tree threws. The present proposal is of great theoretical and realistic significance as it will facilitate the development of reproductive and developmental biology of tree threws.
