中文摘要: 循环肿瘤细胞(CTC)是指自发或因诊疗操作由肿瘤病灶播散进入外周血循环的肿瘤细胞,在一定条件下可发展为肿瘤转移性病灶。研究表明CTC的数目作为标志物在实体瘤患者的预后、分期及复发监控等方面具有一定的临床价值,因此成为癌症研究的一个新热点。相比单纯计数,揭示CTC的分子特征及异质性无疑更具临床和生物学意义,但因为CTC数量极少(每10ml血液只有几个至几十个)且需要单细胞组学技术所以在技术上遇到重大挑战,目前在国际上尚未见报道。本项目将CTC分选芯片与申请人发明的单细胞蛋白组分析芯片加以整合,实现在芯片上连续的肺癌病人CTC分选、操纵以及单细胞尺度上EGFR介导信号通路的分析,从而揭示肺癌CTC信号通路的激活情况及异质性。其中申请人发明的基于抗体微阵列与微流控技术的单细胞蛋白组分析芯片能在单细胞尺度上定量检测EGFR介导的多条信号通路中的关键信号蛋白。这将是首次实现CTC单细胞信号通路分析。

英文摘要: Circulating tumor cells (CTCs) are cells released from primary tumor sites travel through the blood circulatory system, and carried to a secondary site, where they can initiate new tumor growth. Recent studies have shown that CTC detection and enumeration have prognostic and predicitive values, and allow oncologists to monitor patients for disease progression and malignancy relapse. Therefore, CTC study has attracted a lot of attention from the cancer research community. Compared with CTC enumeration, characterization of molecular signature and heterogeneity of CTCs reveals more biological information. However, it meets great technical challenges because CTCs in the peripheral blood are rare events and that single-cell -omics technology is required for such chacterization. This project is aimed to develop an integrated microfluidic platform combining CTC capture chip and single-cell proteomic chip in order to realize on-chip CTC capture, manipulation and characterzation of EGFR-mediated signaling pathways at the single cell level for detecing molecular signatures and heterogeneity of CTCs. The single-cell proteomic chip is based on high-density antibody microarrays and microfluidics technology, allowing quantitatively measuring key signalging proteins in EGFR-mediated signaling pathways.
