中文摘要: 角蛋白是一大类结构复杂的难降解纤维状蛋白质,目前已发现多种能降解角蛋白的微生物和角蛋白酶,但对于角蛋白酶降解角蛋白的过程和机制还很不清楚。本项目以细菌嗜麦芽窄食单胞菌YHYJ-1分泌的角蛋白酶KerF为研究对象,利用光镜、SEM、AFM和生化手段,研究角蛋白酶KerF降解天然角蛋白底物的过程中底物微观结构的变化和酶切位点,阐明角蛋白酶KerF对角蛋白的降解过程;利用生物信息学分析、定点突变和生化手段研究角蛋白酶KerF催化结构域中结合和催化角蛋白底物的关键区域和关键氨基酸残基,以及其C-端PPC结构域在角蛋白高效降解中的潜在作用,阐明角蛋白酶KerF高效降解角蛋白的分子机制;研究典型角蛋白酶KerA对天然角蛋白底物的降解过程和酶切位点,并与KerF比较来揭示不同角蛋白酶降解天然角蛋白的过程的异同。本项目将首次系统揭示细菌角蛋白酶降解不溶的天然角蛋白底物的过程和机制。

英文摘要: Keratins are insoluble fibrous proteins and are difficult to be degraded. Recent studies revealed many microorganisms and keratinases that can degrade keratins, but the degradation process and mechanism are still elusive. This project is to elucidate the degradation process and mechanism of natural keratin substrates by a keratinase KerF, which is secreted by a bacterium Stenotrophomonas maltophilia YHYJ-1. Firstly, we will monitor the degradation process of natural keratin substrates (wool and chicken feather) by KerF using light microscopy, scanning electron microscopy (SEM) and Atomic force microscopy (AFM) and determine the cleavage sites on the substrates with HPLC and LC-MS. Secondly, we will determine the potential function of C-terminal PPC domain in the keratin degradation and the key residues of KerF in the keratin degradation by using bioinformatics and site-directed mutational analyses. Lastly, the degradation process of natural keratin substrates by another typical kertinase KerA and the cleavage sites on these substrates will be studied and compared with that of KerF. The implementation of project will gain insights into the degradation process and mechanism of the natural keratins by a microbial keratinase KerF.
