中文摘要: 药物分子与细胞膜相互作用过程分子层次机理的研究对药物开发、药物作用机制的认识十分重要。本项目将以和频振动光谱为主要研究手段,同时辅以衰减全反射傅立叶变换红外光谱和共聚焦激光扫描显微镜等技术,选择常用的与细胞膜有作用的药物分子(包括水溶性和非水溶性的药物分子),以研究药物分子如何动态地与细胞膜模型体系作用的过程。和频振动光谱拥有独特的表界面选择性、单分子层灵敏度和取向敏感性,可原位、实时地给出药物与磷脂双分子层的动态作用信息,并且可从中获取磷脂分子和药物分子的翻转动力学机制。通过磷脂分子和药物分子在双层膜中的翻转动力学过程,我们可以探究药物分子在膜中取向和排列、磷脂双层膜的完整性和流动性(扩散性、通透性)以及药物分子与磷脂分子的作用亲和力等信息。本研究对于拓展和频振动光谱的应用,阐述药物分子与细胞膜的作用机理,理解药物分子的药代动力学以及进一步设计和开发新的药物等有重要意义。

英文摘要: A molecular-level understanding of the interactions between drug molecules and cell membranes is very important for drug development and the acquisition of the knowledge on the drug's action mode. This project will employ sum frequency generation (SFG) vibrational spectroscopy and other related techniques (such as attentuated total reflectance Fourier transform infrared spectroscopy (ATR-FTIR) and confocal laser scanning microscopy) to study how the selected water soluble and water insoluble drugs interact with the phospholipid membranes from a dynamic viewpoint. SFG has a great advantage of being able to monitor the structure and orientation of the molecules on surfaces and at interfaces with a submonolayer sensitivity. Besides, SFG can provide information on the drug-membrane interactions in situ and in real time, and can also give information on the transbilayer (flip-flop) dynamics of phospholipids and drugs in the bilayer. From the transbilayer dynamics of phospholipids and drugs, we can deduce information on the drug's orientation and packing state in the lipid bilayer, the integrity and fluidity (diffusion and permeability) of the bilayer, and the affinity between drugs and lipid molecules.
