中文摘要: 为了应对日益严重的细菌抗药性问题,寻找新的药物作用靶目标和提高现有药物的药效,我们就一种新的Ser/Thr 蛋白激酶YihE在药物存在下对细菌的保护机制开展了研究。通过分析此蛋白激酶在包括药物在内的不同环境压力下对细菌的保护作用以及影响其保护作用的各种因素,确定了与YihE功能相关的细胞因子及其可能的作用途径,初步探明了YihE在药物存在下对细菌的保护机制,即YihE通过拮抗细菌程序性死亡系统MazEF,减少细菌内由于药物处理所导致的羟自由基的积累,从而降低药物对细菌的致死效应。该研究有助于我们进一步认识细胞与外界环境的相互作用及细菌在药物存在下的生存机制,所揭示的YihE 对细菌的保护机制可以让我们从一个新的角度认识细菌抗药性的产生,为更好地应对细菌抗药性问题提供新的思路和方法。

英文摘要: To contain the increasing development of antimicrobial resistance, explore for new targets, and enhance the efficacy of drugs currently in use,studies was carried out on a novel Ser/Thr protein kinase, YihE, which protects bacteria from antimicrobial agent-mediated cell death. Through analysis of the protection provided by YihE for bacterial cells under various environmental stress, including drug treatment,and the factors that affect its protective effect, we identified cellular factors involved in the function of YihE and its possible pathways. The revealed mechnism showed that YihE provided protection for cells by antagonizing the function of a bacterial programmed cell death system, MazEF, and therefore mitigating stress-stimulated accumulation of hydroxyl radicals that ultimately cause bacterial death.The project led us to a better understanding of the bacterial responses to environmental stresses and and the mechnism of surviving under drug treatment, which would provide new insights on the emergence of antimicrobial resistance and shed light on new ways to contain the development of bacterial resistance.
