中文摘要: 壳聚糖是无毒、生物相容性好的天然阳离子多糖。它作为止血材料、血管支架、神经导管、骨替代物等方面已有大量的研究。褐变降解是影响壳聚糖质量的一个普遍问题。但目前对壳聚糖褐变降解研究围绕褐变程度进行,缺乏对壳聚糖褐变降解物质基础研究,对这些。本项目将建立壳聚糖中褐变产物的分离测定方法,鉴别主要褐变产物;通过降解动力学研究和中间产物研究阐明不同条件下壳聚糖主要褐变降解机理;采用制备液相等手段得到褐变降解产物对照品,评价褐变降解产物对壳聚糖安全性的及其对凝血系统的影响。阐明壳聚糖褐变产物物质基础及其安全性和对凝血系统的影响等生物效应,将为全面认识和评价褐变对壳聚糖的质量和功能影响提供科学依据;为完善壳聚糖的质量控制提供理论和技术基础,同时为壳聚糖褐变产物的进一步研究提供基础。

英文摘要: Chitosan is a non-toxic, biocompatible natural cationic polysaccharide. It has been widly applied as hemostatic dressing, vascular stents, nerve conduits, and bone substitutes. But the browning degradation, a common problem of chitosan, limited the quality and application of chitosan. However, most reports discussed on the intense of browning and few focused on the material basis of browning degradation. The present project is planning to separate and identify the browning products of chitosan, illustrate degradation browning mechanism by studing the kinetics and the intermediates of browning. The safety of every main browning products and its effect to the coagulation system will be further studied based on the isolation of main browning products using prep-HPLC. Identification the material basis of chitosan browning products, explore their biological effects, including safety data and influence to the coagulation system, will provide a scientific basis for the understanding and evaluation the influence of browning products to the quality and function of chitosan. This will also provide the theoretical and technical basis for improving the quality control of the chitosan, and provide information for further study of the chitosan browning products.
